Detection of infectious agents in the respiratory tract

Here we present the respiratory pathogens detected by Labor Berlin in weekly updates. The upper part of the graph shows the positive detections for SARS-CoV-2, influenza A/B and respiratory syncytial virus (RSV). The lower part of the graph shows the results of two multiplex tests. Here you can see how often individual pathogens were found in comparison to the others. The x-axes show the calendar week of the 2024/25 season and the total number of samples sent in.

  • Sysmex XN-Based Evaluation of the Diagnostic Performance of High-Fluorescent Cells From CSF as a Supportive Diagnostic Criterion in Neurological Diseases

    10.04.25 | In a study with nearly 300 cerebrospinal fluid samples, employees of the Department of Laboratory Medicine were able to demonstrate the diagnostic value of the automatic analysis of highly fluorescent cells (HFC) using fluorescence flow cytometry. The automatic cell differentiation, which is carried out around the clock, shows a 95% agreement with manual microscopy and an 80% concordance for HFC. The determination of HFC alone allowed a correct clinical stratification with up to 70% of cases with hemorrhage, inflammation and neoplasia. These results emphasize the potential of HFC as a valuable parameter in neurological diagnostics.

  • TGFβ links EBV to multisystem inflammatory syndrome in children

    20.03.25 | Employees of Labor Berlin supported clinicians at Charité in discovering the cause of a severe inflammatory syndrome called MIS-C that can occur in some children and adolescents after a SARS-CoV-2 infection. The syndrome is characterized by impaired activation of memory T cells due to high TGFβ concentrations. This dysfunction leads to reactivation of the Epstein-Barr virus and an increased inflammatory reaction, which can be reversed by blocking TGFβ. Targeted treatment with TGFβ blockers is being considered as a possible therapeutic option to improve T-cell reactivity and prevent both EBV reactivation and the associated hyperinflammation. The findings were published in the renowned journal Nature.

    Advanced disease and CD8+ TEMRA cells predict severe infections in multiple myeloma

    28.02.25 | In a prospective study, staff from the Departments of Immunology and Haematology & Oncology, in collaboration with clinicians from Charité, investigated risk factors for severe infections in patients with monoclonal gammopathy of undetermined significance (MGUS) and newly diagnosed multiple myeloma (NDMM). Almost half of these patients suffered infections within the first year after diagnosis. Particularly advanced disease stages and increased concentrations of CD8+ TEMRA cells (‘terminally differentiated effector memory T cells re-expressing CD45RA’) were significantly associated with an increased risk of infection. The developed risk assessment model PREDICT-SID, which integrates clinical, laboratory and immunological parameters, was able to identify patients with a sensitivity of 93% and a specificity of 80% (AUC 0.96) who are at high risk of early severe infections and may therefore require more intensive preventive and therapeutic measures.

  • A candidate reference measurement procedure for the quantification of α-synuclein in cerebrospinal fluid using an SI traceable primary calibrator and multiple reaction monitoring

    29.01.25 | Neurodegenerative diseases such as Parkinson’s disease and Lewy body dementia show α-synuclein aggregation in the brain as a common key feature. Due to considerable problems with the standardisation of previous α-synuclein measurement methods, caused by the insufficient specificity and measurement accuracy of the antibodies used in conventional immunoassays, members of the Department of Immunology have developed a new reference measurement method (RMP) in an international initiative. This method, which is based on an internationally standardised (SI) calibrator and the use of liquid chromatography-tandem mass spectrometry (LC-MS/MS), enables reliable and standardised quantification of α-synuclein in cerebrospinal fluid in order to ensure comparable measurement results for patient care worldwide.
